Anyone know if it is possible to connect two routers using a 'crossover x.21 cable' (if there is such a thing)?

Reason being to test leased line configurations without actually having a leased line...

I was told this was possible if one router acted as a clock...

Any thoughts gratefully received!

Yes, what you describe is possible provided your router can act as a DCE and provide clocking to your "circuit". You'll need either a cable specially made as a cross-over or you can also buy a standard male v.35 DTE cable (used for connecting a router to a CSU/DSU) and a female v.35 DCE cable. You then connect the two v.35 connectors together and add appropriate clocking commands to your router. The router on the DCE end has to be the router which will provide clocking for the "circuit". You don't mention the type of router you are trying to use, but if you have Cisco routers the following is all you need to add to the interface configuration:

clock rate 2000000
(or whatever clock rate you want in bits-per-second up to 4000000 on a 2500 series for example)

Hope this helps.
