Windows 2003 Fileserver

We are having an issue with deleting files that are located on our fileserver. The fileserver is a Windows 2003 R2 Standard edition server. We get an error that the file is in use but it is not opened by anybody on the network. We have used unlocker and process explorer but they did not work. If we reboot the server then we can delete the file. We just don't want to keep on rebooting the server each time we have this issue. Anybody out there also have this issue? Anybody know how to fix it?
 
I have already called our Backup software tech support and we ruled out that the backup software is causing the issue. We also disabled and stopped all the AV services but we still have the issue. Thanks for the info though.
 
They only have a mapped drive to the shared out folders on the fileserver but not the files themselves.
