Vlans are easy enough to setup, just create the vlan and assign the ports to the vlan. If you are assigning more than one port to a vlan you will probably have to use tagging or 802.1q as it is known.
As for spanning tree, you just enable to disable spanning tree for the port and not the vlan. In the case of a 3300 switch for instance you need to enable STP for the whole unit and then go into the port option and and enable or disable it. Note that the later codes have FastStart STP which is similar to Rapid Spanning tree, this cuts down on the convergance times.
