Is there a way to busy ISDN30E trunks on a Mitel 3300 to prevent incoming calls coming into site for testing purposes, as a manual busy of the trunks is not recognised by the Network Provider. ( apart from dialling out all channels at the time or getting the Network provider to busy them )?

The only other way is to disconnect the link
Trunks that are ManBusied will not receive incoming calls whilst in this state.
