Is it okay to plug a 12x0 trunk module into a 6 port fiber expansion card without powering the ICS down?

yes you can do it. disable and enable the module, it should work. start at the top of the fiber card.

You should not have to enable or disable unless an issue.

Should I make the fiber connection before applying AC power to the trunk module or make the fiber connection with the trunk module already powered up?

I don't think it matters, it wont hurt either way but best to power up last.
