Where can I add mailbox rights for one user to access another users mailbox? In 2003 it used to be under that persons AD record in one of the Exchange tabs.
 
You would use the Add-MailboxPermission cmdlet.

Add-MailboxPermission "mailbox name" -User "usertowhomyouwanttograntrights" -AccessRights: FullAccess

Just you run this powershell cmdlet right from the Exchange 2007 running that runs ESM or could you have done it from one of your DC's?

You could have run it from any workstation in the domain that has the management tools loaded on it, which includes the PowerShell. You can run the management tools on a DC if that's your goal. The tools come in a 32-bit version.
