Hi,
Start up Exchange system manager,expand the server folder until you see the Queues. click on the Queues and on the right hand pane double click one of the connection queue names to open "Find Message" dialogue box, click Find Now button and on the "search results" window selct all the messages you to delete, right click then select delete.

To delete ALL outgoing mail in the Queue at on time go to
"\Program Files\Exchsrvr\Mailroot\vsi 1\Queue" on the drive containing your Exchange program and delete all the files in the "Queue" folder.
Handle your Exchange as it should, like youlie explained, NOT by manually deleting files, EVER!

Youlie's response contains the correct information to delete messages from the queue in normal situations and was sposed to be the first part of my response.
The portion of my response needed to be prefaced by the following:

In the event of an open relay, and the queue contains thousands of messages and hundreds of queues you can delete all the messages in the Queue folder at one time. Stop the SMTP virtual Server before continuing.
