Exceptions list blocking access to site

I have users who are trying to access a site behind a portal. Behind the portal they are able to access an applications page that allows them to choose their app. However, when they click on any link to an app. they will get a Page cannot be displayed error. If I go to the Internet Options -> Connections -> LAN setting -> Advanced and remove the exception for that site, they work fine. But, the error has started coming back for several users because each time they reboot or log back in the exception comes back. I have the exception on my list, yet I work fine. Any ideas what is causing this and if I can fix the problem once and for all?
