Database running slow

Are there any simple checks i can run to find out why a database is running slow. i checked the server and it is really badly fragmented so i am currently running a defrag on it, but i am wondering if there is anything i can run at a db level.

i've checked the server that holds the database devices, and the drive is very badly fragmented, i have tried to defrag the drive but it will not defragment the large database Device files, so the drive remains fragmented?
