You can't transfer directly from one machine to another using a normal USB cable.
If you really want to network using USB you can find out more about it here:
I would recommend 'proper' networking though, if both machines have ethernet. You use something called a "crossed" or "patch" cable - they're slightly different to normal ethernet cables. They're cheaper than USB networking.

Just a warning - it's possible to get hold of USB cables that have the same flat, wide connector (i.e. the one that plugs into your PC) on each end. Unless these specifically state that they are USB networking cables do not use them to connect two computers together. You can cause serious damage if you do this.

Yes, install both NIC cards. Get them active (Should happen automatically after a reboot). The cable you will need is called a cross over cable. Connect both the laptop and desktop. From WHERE you are transferring files, make sure to SHARE the folders and you'll be able to move files!!!
Or instead of a cross over cable, you'll need an inexpensive hub and 2 regular LAN cables. Share folders as in the above directions and your all set.

OOOPS.. should have asked, are you SURE you don't have a port on your laptop labelled com? That's the serial port.
You might also want to look for a program called laplink. It's purpose is excatly what u are looking for and SHOULD include the proper cable(s).

In that case you don't need an ethernet card for the laptop. A LAN port is the same thing.

If your PC doesn't have a LAN port (check first!) then you need a network card for it. They are very cheap, in the UK I wouldn't pay more than 20 UKP (about 30 Euros).
You then need an ethernet cable, also known as a network cable, but you need a special kind of ethernet cable called a 'patch' or 'cross-over' cable. They are very common and should be cheap.
Follow the instructions for installing the network card in the PC. When it is all set up connect one end of the cable to the LAN port of the PC and the other end to the LAN port of the laptop. Go to the Control Panel of one machine and run the 'Network and Dial-Up Connections' item. Double-click 'make new connection' and follow the wizard - I think the one you want is something like "accept incoming connection".
My knowledge is a bit sketchy after that point because I haven't done this for years Other people here should be able to help you if you get stuck though.

Cat 5 Cable (Crossover)
Crossover cables have 2 wires crossed over on one of the RJ45 connectors. It would be even better if your laptop had wireless and you just installed a wireless card on your PC. That may be the most expensive route, but you dont need wires.
You should be able to find a network card pretty cheap for a PC. Since most new motherboards have Integrated network cards many old computers that are no longer used had network cards in them that are not needed now. So there should be a glut of useless network cards just laying around.
Another option is a USB to Ethernet Converter. I have seen small devices like this for wireless networking. The concept being that you dont have to open the computer.
Here are two examples of one for wireless and one for Wired. These are just examples.
