This is actually a lot simpler than you'd think.

Since you said you've set it up on your registrar's DNS panel, I'll skip that part.
Short answer is you need to create a new website in IIS ( right click on the Web Sites folder and select New|Website) Give it a friendly name on the first screen, and click next. This next screen is where it's at. For me, anytime i use more than one site, i have to change the IP address from "All Unassigned" to the actual IP of the machine. If you do this, do it on all your sites. Port 80 can stay the same unless you need something else. Host headers. This is where you type in the DNS name of the site. in your case video.mydomain.com is what you want. All this is doing is saying that if a request comes in with this header, then direct it to this newly created site.
Simple as that.
Also if you've already created the site in IIS, right click the sight and go to properties, select the website tab if it is not there by default, click on advanced, and you see a list of host headers this site will respond too. delete what is there if you don't want it, and click add. From here it's the same thing. IP address, port and header value (video.mydomain.com)

Let me know you need help!
