Client not connecting to SQL Server instance

Originally there were two applications on the same SQL Server. The decision was made to create an instance of SQL and move one of the databases to the instance. This was done but the client could not connect. So the data from the other database was move to the instance and again the client could not connect. What are the various solutions to resolve this problem? TIA
 
Sounds like a security issue. Are both servers set up the same and do the databases have the same users, etc.?

Both servers?"...only one Win2k server with two applications installed and a single SQLServer installed. Want to create an SQL instance and segregate the two databases.
 
The data was moved to the instance which the client cannot connect with, the client was able to connect with the original database out any problems.
