backup exits with status code 1

Backup exited with status code 1 (partially succeeded). I have an image in the database corresponding to this backup. If I see the problems reported for this job ID, it displays a warning message saying that a particular directory could not be opened (WIN32 5: Access denied).

Since a backup image is available, can I safely assume that except this file, everything else was backed up properly? Or is it that the backup process stopped with this file and didn't go beyond.

Yes, the backup is a good one except for the file(s) for which warnings are raised.
Additionally, You can also expect return code 1 for good database backups, if you are using scripts. Safely ignore those.

Yes the backup was successful except the that file.
Find out why the file was not backedup. File might be open.
