Alter single file with CACLS.exe?

I'm trying to alter the permissions on a single file in a folder but as it is inheriting permissions so it doesn't seem to let me. Is there a way to block inheritance for the files with CACLS.exe as well as modify permissions?

You need to modify the ACE as well as the ACL permissions on the file.

I was under the impression that ACE's we're only used with the deny permission, am i wrong here?
