Thanks for your input - I'd gone ahead an rebooted the server last night before waiting for replies, and the problem has dissapeared.
I still have no idea what caused it tho. I'm guessing the it's to do with the network adpater getting all crossed up rather than a DNS/AD issue.

Forgot to add, when you attempt to browse the network on the domain
controller machine, i get the following error pop up:

What OS are the macs running? If you're on any version of OS X, they should be able to browse the windows network at least. Gimme some more details, I might be able to help you out.

What I mean is, I added @<mydomain>.com as the default email domain, so all the default smtp addresses for each mailbox on the server are blah@mydomain.com.
The firewall does nothing to the mail, just forwards it.
We've got no problem sending mail out, just recieving from certain domains...

Yesterday I put our new exchange2000 server in, and everything is working fine, apart from incoming SMTP mail from some domains - it gets bounced with something along these lines to the sender:
