Are you using Symantec AV? This can cause the problem - you need to exclude .vsp files from the realtime scanning.

Sounds like something might be running in the background that causes the problem. Does it always reach the same set of files/directories in the backup? As an easy first test try moving the time the job is scheduled for by a couple of hours in either direction and see if this makes a difference...
