Are you sure its a problem with Exchange?? Firstly ensure the Exchange Services are still running at the weekend.. if so, perhaps the messages are actually being processed but being queud up somewhere else?? Firewall?
Frequently on our Exchange 5.5 server, under mailbox resources - the NT a/c associated with many of the mailboxes is often the ADMINISTRATOR. The ADMIN isn't actually the primary account associated with these mailboxes but my understanding is during the course of the "backup" procedure...
