The 2420 and 5420 is digital and only require a single pair to operate. The 4620 provides a back lit display so do these handsets require either the power injector or can they get power from the IP office itself or from a switch that provides POE? Do the I need any additional licenses to install the IP vs the Digital handset or a different card installed in the IPO?

The 4620 is an IP phone so it does not connect directly to the IPO. It requires a POE switch, or brick.
You need CAT5 or better wiring to connect this set, layout according to T568a or B layout.
Yes you most probably need to get licenses if you never had IP phones working.
What you need is partly depending on your current software release.
Also you need to properly set up your LAN for carrying voice.
Based on your questions, I would advise you to get in touch with an Avaya partner in for a quick scan,
which you would need to contact anyway, if you need to purchase licenses.
