whether EPO2.0 support to upgrade/update to the latest virus defintion

I have a EPO 2.0, I don't know whether it support to update/upgrade all client PC to the latest antivirus definitions (e.g. 4221).

Yes it will. I would recommend that you upgrade to 2.5. Less problems than 2.0 :)
