I have a virtualized time & attendance application that is going to need a access to a modem. Our timeclocks are all IP based except for a remote building that has no other connectivity but phone (blech) for the near future. My host is a HP Proliant DL380 and I appear to have a serial port available.

Am I going to be better off using a serial modem or a USB modem? Which will be easier to configure/more stable under ESXi? (The client OS is Windows 2008R2.)

You can map either the serial or the usb to a virtual machine. But keep in mind, this will tie the VM to the host. To give the VM the ability to vMotion to another host or to HA to another host, you will need to ensure there is a modem and a serial port on the other hosts as well.

Don't think there would be an advantage between USB or serial.

You can also look into TCP modems.

I thought about the host tying. I don't think I have any choice here.

Unless I'm not searching the proper terms, I think a TCP modem is the opposite of what I need. The remote building has no communications except an old fashioned phone line.

We used a serial to ethernet converter to connect a laptop to one of our virtual machines. That way the vm could migrate between various hosts without losing it's modem. The brand we used was Digi, but there are a ton of companies making them.
