I have a user that claims that she only can send e-mails to a specific domain if the e-mail has "Re:" in the Subject line. Is this even possible? I haven't seen any proof of this, except the the recipient of that domain sending the user e-mails saying that they aren't getting her e-mails that she was supposed to be sending them...

Yes it's possible. A whitelisting scenario could cause this. For example, if the recipient is set to only recieve mail from whitelisted senders unless it's a reply to their message, this could happen. In fact, have you noticed that a lot of spam comes in with Re: in the subject line? - primarily to get around filters like this.
