Unavoidable jerks in adobe premiere 6.0

I've been making a music video in Adobe premiere version 6.0 but in my editing i have experianced a VERY annoying problem.
In just oine part of my video the audio and video both jerk for about 2 seconds then carry on- there is no gap in between where the clips meet and i have tried cutting the edges and putting back together with no avail.
I've also tried putting the clips on differant video lines
and overlapping but this doesn't help either.

I'm really crap with premiere- I also can't use transistions, i put them in teh transistion line and follow all teh instructions so i play it and nothing happens.
Anyone know why?
Help would really be aprecaited.

You must render the project before the transitions appear.

The 2 second glitch sounds like it is in the raw footage, maybe try re-capturing it.
 
Try dragging the yellow work area bar just to the edges of the problem area. Then export that small clip to video. Play back the result and see if it's okay. If it is, use the clip you just made to replace the problem area in your edit. Jerks like that can be from your hard disk trying to do too much too fast, and a single clip solves this problem.
