I have some footage of a bride carrying a bouquet. My goal is to isolate the flowers and have them appear in color while the remainder of the frame appears in b/w. The clip is approx. 20 secs in duration.

I do this all of the time in Photoshop with stills but I am finding AE5 very difficult.

Someone... please help

Animated Masks.

You'll need to find a tutorial on masking first. That will explain how to create the mask.

Then, set keyframes for the MASK SHAPE over the duration of the clip. You only need to set the first keyframe, the rest will automatically fall into place as you edit the mask shape.

Last, duplicate the video to a layer BELOW the masked layer.

On the mask layer - no effect.
On the layer below - Desaturate.
