I am assuming you are asking how do you configure the clients to pull updates from your SUS box?
There is website called that has a good FAQ and tutorials on how to configure the clients.
When we deployed our configuration we used group policies thorugh AD. The SUS Whitepaper from Microsoft explains this rather well.

We also use group policies for our SUS. Under Computer Configuration\Administrative Templates, add a the wuau temmplate. That will give you the Windows Update piece of the GPO. Then set it to point to your SUS website, set a time, and set the install options and you are good to go.
