User A: was granted access to a secure site. When the window poped up prompting them for a user name and password, they checked the box indicating to save the uname/pwd. They however, typed the wrong uname/pwd, and cannot enter a new uname/pwd.

how do you Uncheck the box or get it to pop up again?

 
Clarification: This would be the "Enter Network Password" window
 
Assuming that your user is using IE....Tools, Internet Options, Content tab, Autocomplete button, Clear passwords button.
 
Well that clears the autocomplete passwords, but not the password if a "network password" window pops up. I'd include an image if I could.

1st line - User Name
2nd Line - Password
Lower Left corner - Check box "click here to save password"
I need to find a way to uncheck that box and reset the user's password.
 
What operating system are they using?

Yes, this is related to the operating system, not the browser.
