Is there a way of quick way of assigning and unassigning a skillset for a group of agents in AACC6.

Yes, go to 'Contact Center Management' and open menu on the left and click on 'Skillsets'. Select the proper skillset, 'Unassign' and send.
 
Just in case this is a regular occurrence you can also setup predefined and scheduled assignments in the same menu area.
