I want to draw a rectangle in Illustrator, then bisect it with a line. How do I fill in one side of the now bisected rectangle with one color and the other side with another color?
 
Make sure both the line and rectangle are selected and then click 'divide' in the pathfinder palette. You will now be able to select either part of the rectangle seperately with the direct selection tool and give them individual fill colours.

TIP: You can group the two objects to make sure they dont get seperated and still select them individually and change their fills even after they are grouped.
 
try using knife tool ,

divide the rectangle into two separate objects

then fill both parts with different colors of ur choice
