Setting up VPN from scratch

Hi!
I'm a little confused about VPN. I have a situation:
LAN at work with one PC running Win2KPro with 2 NIC (and acting as file server). One NIC for the LAN, the other for Internet via Cisco Router (ADSL). I have a static IP for WAN. Is it possible to connect to the LAN from whatever PC connected to Internet, directly through ex. IE5? What URL should I use from the client? Is it maybe better to set up a ftp-server with IIS?
