I have a exchange 2003 and I use outlook 2007. Here is the questions: How can I have my assistant send emails on my behalf? Is there any configuration that needs to be done on the server?
Can be done either way. There is delegation rights within Outlook or you can set "Send on Behalf" rights in the server under Exchange General tab->Delivery Options button in your user properties page. Either gets the job done.

Thank you much. I made the changes on the server. Now how within outlook can I use this option? It shows only my account. Do I have to do something extra?
When the person who has the rights sends e-mail, they need to enable the FROM field (usually under View->From: in the actual e-mail). They would simply populate the FROM field with your userID/mail account. Exchange should then allow the e-mail to go through, and the recipient will see in the from area "From [sender] on behalf of [user]"
