We are also planning to go from 4.2 to 6.0 and our vendor hasn't said it will be a problem. That said we will likely be going from a distributed (10 @ 4.2) to a centralised (1 + 1 for failover @ 6.0) architecture; we won't be upgrading the boxes, but forklifting in new ones.
Yes, you can go directly from 4.2 to 6.0 without upgrading to 5.0 first. There are higher server hardware requirements, though, and it will use Windows Server 2003. Assuming you are migrating to a new server, you will have minimal downtime. If using same server, you will need to completely reload (don't upgrade from 2000 to 2003).
Are you also using Web Client? There is a way to migrate your Active Directory, but it does require a separate server. Otherwise, if you can just reconfigure, you might be ahead of the game.
The "classic" client is gone. But Web Client is greatly improved, and the Scripting component no longer requires Terminal Services. (yeah!)
