Presumably you are using the save for web function to convert a vector Illustrator file into a raster GIF? GIFs are based on pixels and so therefore cannot be upsized without those pixels becoming obvious. If you need to keep clarity in a large version of your artwork in a pixel format, start off with a large version of the AI file before converting it to a web file format.
