Can I use variables in filters and query items?

Do you mean parameters? You can use them in a filter and in a report expression..

No, I actually wanted to create a variable. This doesn't appear to be possible though, since Report Studio variables are mainly used for displaying, or not displaying content on the report.

Yeah right , as in boolean variable. However these will either default to true or false (1/0)
The logical expression behind the variable itself should be able to serve to build a filter?

There is a place for you to do these as chosen when you chose the boolean, but you have to set it up properly. Depends on what you need to do. But, it can be done.
