re-label a live system disk

Does anyone know if you can re-label a live system disk in the sense that there is space still on the disk that can be assinged to create a new partiton. Do not see any reason why you could not, my understanding is that the label info is only read at mount, but a colleague seems to think it is a no no, anyone know why ??
