I've used it many times to migrate tens of thousands of mailboxes. It's not the most simple of tools, but it generally gets the job done. Quest has started to really make some positive changes in the Coexistence Manager tool as well, that removes the requirement for Transporter Suite.
You can run multiple migration workstations to increase velocity if your network and Exchange environment will support it.
Your Notes, Quest, and Exchange environment should be on the same LAN segment, not separated by WAN links.
BTW - you'd be better off posting this in the Exchange 2010 forum, forum1733, as you'll get a more dedicated audience there.
