Is their a global setting so a key system can have privacy
on all lines on all phones.

nope, either assign a privacy button to each phone, or dial (Feature 31) or go off hook on the intercom and dial (#31) at each phone.

Will #31 work if you copy the extension to another?

you can copy the privacy button to other simialar telephones, but you won't be able to copy the enabled feature to other telephones.....I hope that made sence.
