Permissions on sites disappear and come back......sometimes 2007

We have a Sharepoint 2007 farm that permissions were working correctly. We have been seeing an issue where teh permissions are not being applied to certain sites but the correct permissions will return after some time. The first time we saw the issue was on a team site where users who had teh correct permissions could not upload documents. After 30 minutes this was working correctly again. This has happened a few times since the issue was first reported. We have since seen this happen on other unrelated sites as well. Now we have a site the permissions are not being applied on again but they have not returned as well. When I log in with the System account there are no elevated permissions available. When I go to the site permissions page I can view the permissions and the sharepoint groups but I cannot edit any of them.

This has been happening for about a week and I have not been able to locate any possible cause for this. We have a single server that hosts everything but the contentDB and the SSP DB. These are stored on a seperate DB server with proper space. The content DB is 150GB in size which has me concerned and I planned on splitting the database in the future since I believe the recommended size is less than 100GB. I would not think this is an AD connection issue as the other sites are working properly and elevated permissions are there with no issues. No major errors on the servers.

Sharepoint SP2 and updates were applied about a month ago. Are there any known issues with any patches that may relate to this? Has anyone seen any issues like this? Could the large content DB be causing this issue?

Any help is greatly appreciated.
 
The Sharepoint farm now allows administration of team sites but not the main portal site. This has been the case for 5 days now and permissions have not returned. I have verified as much as I can with the system account but it too cannot administer the main portal site. Any idea?
