I was looking around on the net and found "The art of Assembly language" and I'm not real sure if I should read the 16bit of the 32bit book. Any one have any suggestions? Can you program stuff for MS-DOS using 32bit ASM? Also, does the book reqiure any knowlegde in any other programming languages? Thanks for the help...

All current processor is 32bit, and 64bit is already came out. So you should read a book for 32bit. But for the basic, it is helpful to read 16bit. So my suggestion is to read both :)

If you mean 32bit for real DOS, I really don't know (never tried that). If you mean for Windows DOS console, then yes you can. To make 32bit program for windows, you also need to know about Windows API programming.
