Newbie SharePoint Designer Question

I have a SP2007 site set up in an AD domain with host headers and a DNS alias: The site was created with the Collaboration Portal template. On the home page I am trying to resize the Middle Right and Middle Left Web Part Zones.

I've fired up SharePoint Designer and connected to the site via the url listed above. I open Pages/Default.aspx. I see the master page open up and get what appears to be the home page. I've made my sizing changes, saved the page and checked it in. However, the changes do not show up when I open the site in IE. I also tried publishing, however, the site shows up as the local site and SPD wants a different url for the remote site, so that's not the method.

What incredibly simple and obvious step am I missing?

I tried this 3 or 4 times before posting, but later trying again it worked correctly. Don't know what changed.
