i want to force specific analog extension use specific route for external call like route 6.

First of all, you have to initiate dest for the route 6, then initiate again the same dest but this time with adding cust parameter.
For example cust=1;

Secondo, you have to go back to the Analogue extension settings and make change command to add cust with the same value of dest cust.

You can use traffic_matrix but I don't recommend that specially in a huge systems.
