When the MP card is replaced on a 2000(2) system can the same 48 PORT software be used or is it married to the original MP card?

Yes,
You can reload the software and reregister it. If it won't reregister call NTAC and tell them what happened.

No you CANNOT load the original software onto a new MP. All software is registered to the specific MP serial number.
When a replacement MP is ordered you should give NEC customer Service the serial number of the one you are replacing and they will burn that number to the new one so that you can use the existing software at the site.
If you try to load the existing software to a new MP it will not register and your only option is the temp activation code in CM F83 which is only good for 120 hours.

I am sorry if I said you could reload it on a new processor. I have never actually had to replace a working 2000 processor, but I have got a used processor and loaded new software only after contacting NEC to get the new software. Sorry about that. Anyway I hope you got it up and working again.
