I've been working with the new .adp file format in Access 2000 for some while and it's like trying to learn database design all over again. I want to use the easier .mdb file format but I've read that it has the capacity of only 2GB, as does MSDE. I will eventually move to SQL Server Enterprise Edition, so my question is will this off-set the 2GB capacity of my .mdb file

I don't know what you mean by offset, but if you use Access as a front end and SQL Server Enterprise on the back end, you no longer need to store in your .mdb all of the tables that you have on SQL Server.
