Locking out the C drive?

Is there a way I can lock out a specific user from viewing anything on the C drive without adversely affecting the system, because I tried going to security settings on the drive and changing them, but after I do so Word refuses to load, it goes through that "installing office xp" screen (like when you first run it) but cannot complete it while the hard drive is locked. Even if you unlock the drive, let it run, then lock it, it still doesnt work. Is there another way to do it?
 
Can't you set the access rights on each subdirectory to only allow those you want to access it..

What did you change the security settings to? (just Traverse folder / execute file)
