Keep PCs on 24/7? When to run AV scans?

Is it a good idea to keep your PCs on 24/7? The way I see it, that would allow for anti-virus scans to run off-hours, making virus definitions for people current when the come in.
Also, how often should one run AV scans if you have real time scanning happening already?

Any questions/comments/suggestions would be appreciated.

I have it setup to do a full scan the 1st of the month at midnight. I do a Wake on LAN to the computers so the scan runs that night/morning without the need to keep the computers on all the time (or tell users to keep them on which doesn't cut it).

I'm also curious what other people have set for their schedule in regards to your point.
