I have recently installed ISA and have an application on a PC behind ISA that needs to run ftp updates on a daily basis.
I have been told I need top allow 'port 25' open on the Firewall.
I am not familiar with ISA and so far have managed to duplicate the default ftp services that use 'port 25'. I have left the original ftp services active and on the default 'port 21'

Can anyone help me please?

That application sucks. Its using port 25?!?! That's reseved for SMTP.

Essentially you can open any port you want. If the application uses that port then it will work.
A port is a port. If an applicaiton is desinged to use that port then it will. If the port is open, then it will connect, and everyone is will be happy...until you try to put up an email server.

Does this clarify things at all for you? If not, tell me what you're missing, and I'll do my best to help.

Thankfully on this occassion ISA is purely for web browing and ftp updates no SMTP is required.
Does ISA configure SMTP automatically? If so do I need to unconfigure it? (if so how) or can I free up 'port25'?

from a DOS promp I need to be able to:
ftp> open 62.231.140.82
connected to Globscape (62.231.140.82)
This connects me to the ftp server in question, if I get this far then the application will update.

This works from the ISA server PC but not from any of the PCs behind the ISA server.
I need port 25 available, open and ftp allowed on that port through the Firewall.

Thanks for the help

Not sure if this helps but I had the same sort of problem when trying to ftp to a web server. To get round this I went into network settings for TCP/IP on the client machine and added the IP address of the ISA server.

sorry. forgot to mention that I added the IP Address of the ISA server as the gateway (worked on both w98 and w2k).

Like adeleaf said, you need to have ISA as the gateway, or at least have the route to the ftp server in the client pc's routing table.
Generally, if you can do it from the ISA server you can do it from the clients, as long as there is not a routing issue.
