Is resolve.conf read at boot time or real time

We're amending our network and one of the things which is changing is the list of DNS servers

The simple fix is to amend /etc/resolve.conf - do I need to restart the networking service as well?

I'm fairly certain it's read in nearly or literally real time. You can test this by running a ping by name with a valid resolv.conf, then alter/blank resolv.conf and try the same ping immediately without restarting networking.

Restarting the network service is really a pretty rare event under normal operations, IMHO.
