Is there a way by which more that 255 fields can be defined in a table or query in access 2000.

Nope.
If you really need more fields to store data, you'll have to create two tables and set a 1-to-1 relationship between them.

But you cannot retrieve more than 255 fields in a query.

Sad, but true. I have never needed such a number of fields, but I don't like limitations...and this is one of them.
