I have a POP3 connector which polls individual mailboxes from an ISP and then delivers messages to that users MS Exchange mailbox. How would I go about creating a rule for all messages that enter the server in this way. I want a rule so that if a message contains the word Spam in the subject header, it moves it to a particular mailbox for screening.

Don't think you can do that with MAPILab but you can do some things using the IMF.
