I want to have two direfernt email accounts going to one exchange box

Exchange 2k btw

1. xxxxx@email1.com
2. xxxxx@email2.com

First one is already set up is it simply a matter of telling ISP to route all of second one to exchange box and setting up the users accounts in AD?

Yes - make sure that exchange is configured to accept both domains too - and also that the new domain is included in the routing rules (if you are using this method to stop external relaying)
