I need help with COMPANYWEB (ASPX. issue)

HELP! I'm getting the error msg. below when I try to access our company web.It seems that the borwser is looking for /_layouts/1033/settings.aspx. So I checked and found it in the 60 BIN folder, (just to see if was there--and it was) question is how come the broswer (or IIS) isn't seeing it? And how can I remedy this situation? I'm pulling my hair (or what remins of it) out trying to figure this out.
