How do I set up a 2 color file in PS 5?

I want to design a cover for a brochure using two colors (blue and black) which I will then flatten and save as a tiff so I can place it in Quark. If my document is a CMYK, do I fill the Magenta and Yellow channels with white (or select that channel and delete the color in it) so when I go for film output I will only get film for the black and cyan plates?
 
You could set your image mode in grayscale, after that set your image mode in duotone, select your color in pantone color and save in eps. eps format work in qxpress.
 
The image you are looking at is not just black and red - it is many shades between as well. First open the image that you want to work on, save it as a new file (so you keep your original intact) and then choose Image > Adjust > Desaturate. This will turn it into a greyscale image.

Next doubleclick on the layer (which will be named background) and rename it to anything - this makes it a normal layer.

Add a new layer and drag it below your image. Fill this layer with the background colour of your choice (red in the example given). You won't see this colour yet but here is the cool bit - select the top layer (with your image on it) and in the layers palette change the blend mode to multiply. There you have it!

Play around with blend modes to see the different effects they have - they are one of the most useful features in PS

Also, you can start with the Grayscale=>Duotone route, then change to the custom color channel route and intensify/edit certain areas for more control, saving as DCS 2.0 eps (placer eps plus two channel epses.) You can force the intensity of the color to increase over the center of attention, etc. Check out the manual for variations, etc.

I've seen that give a little extra dimension in Duotones if you don't go wild on it. (But maybe on your project, you'd like to...)
